Characterization of Glioblastoma Cancer Stem Cells Sorted by Sedimentation Field-Flow Fractionation Using an Ultrahigh-Frequency Range Dielectrophoresis Biosensor

Cancer stem cells (CSCs) appear to be an essential target for cancer therapies, in particular, brain tumors such as Glioblastoma. Nevertheless, their isolation is made difficult by low content culture or (<5% of the tumor mass) and essentially based on use fluorescent magnetic labeling techniques, increasing risk differentiation induction. The label-free separation methods sedimentation field-flow fractionation (SdFFF) promising, but it becomes necessary consider a coupling with detection characterization method future identification purification CSCs from patient-derived tumors. In this study, we demonstrate first time capability using ultrahigh-frequency range dielectrophoresis fluidic biosensor detector. This implies important methodological adaptation SdFFF cell sorting new compatible carrier liquid DEP buffer (DEP-B). After sorting, subpopulations derived U87-MG LN18 lines undergo biological characterization, demonstrating that DEP-B liquid, sorted specific characteristics: F1 = differentiated cells/F2 CSCs. These presented high-frequency crossover (HFC) values similar those measured standard (around 110 MHz) CSC 80 populations. appeared promising solution development online integration these two complementary separation/detection technologies.
